罗格列酮对 3T3-L1 脂肪细胞增殖及胰岛素抵抗的作用

要】 Objective : To investigate the influence of rosiglitazone on proliferation of 3T3 -L1 adipocytes and insulin resistance. Methods 3T3 - L1 preadipocytes were cultured, the proliferation of 3T3 - L1 preadipocytes was detected by MTT method. Insulin resistant 3T3 - L1 adipocytes cell model was induced by dexamethasone and the change of glucose concentration in cell culture was determined after rosiglitazone treatment. Results In the high glucose DMEM culture media, MTT method showed that the absorbance at 570nm of 3T3 -L1 preadipocytes were increased and that of 3T3 - L1 adipocytes were decreased. rosiglitazone significantly increased glucose consumption in 3T3 - L1 preadipocytes and adipocytes culture with a concentration - dependent effect. Rosiglitazone improved the sensitivity of 3T3 - L1 adipocytes to insulin. Conlcusions Rosiglitazone can promote the proliferation of 3T3 - L1 adipocytes and inhibited the proliferation of 3T3 - L1 preadipocytes, rosiglitazone can significantly improve insulin resistance induced by dexamethaone.% 目的:探讨罗格列酮 对脂肪细胞增殖和胰岛素抵抗的影响.方法:培养 3T3-L1 前脂肪细胞。用四甲基 偶氮唑盐( MTT )方法检测 3T3-L1 前脂肪细胞及脂肪细胞的增殖情况,同时采用 地塞米松诱导 3T3-L1 脂肪细胞建立胰岛素抵抗模型,检测罗格列酮对细胞培养基 中葡萄糖浓度的影响.结果:在 DMEM 高糖培养基中。罗格列酮可促进 3T3-L1 脂肪细胞增殖,抑制 3T3-L1 前脂肪细胞增殖;使 3T3-L1 前脂肪细胞及 3T3-L1 脂肪细胞葡萄糖消耗量增加;罗格列酮能明显增加胰岛素抵抗 3T3-L1 脂肪细胞培 |
